LABORATORY SAFETY PRACTICES
Following safety measures should be taken in the laboratory or safety measures in the laboratory:
1. We must always remain alert and cautious while working in the laboratory. Careless handling of opportunist
and specimens can cause serious injury and can be dangerous to life.
2. We should know the procedure of experiment and be aware of potential bargards.
3. We should know the location and use of emergency equipment in laboratory accident.
4. We should avoided inhaling gases.
5. We should protect the eyes with goggles while handling fumes.
6. We should not pure water in to acid during preparing solution’s.
7. Prescribed methods should be followed for hitting and mixing.
8. The gas burner should be turned off before leaving laboratory and inflammable substances should never be
kept near the flame
9. A burning match stick should not be thrown in to waste basket.
10. We should safe our self from laboratory acquired infections. Eg, hepatitis, tuber Calais.
11. We should prevent self injury during collection of specimens.
12. We should wash our hands thoroughly after collecting specimens.
13. All chemicals, re-agents, solvents and solutions should be appropriately labeled and stared.
14. The following should be available in the laboratory:
Chemical fume hood for handling corrosive, carcinogenic, toxic and radio active substances.
Emergency showers and eye washes.
Fine blankets.
Proper temperature control and ventilation.
Proper waste containers.

ESTIMATION OF SERUM TOTAL PROTEIN BY BIURET METHOD.
Principle: in alkaline solution cupric ion react with protein & form a purple color complex. The absorbency of these complexes is proportional to the protein concentration in the sample.

b. Regent: (Biuret regent)

copur sulfate
k-iodide
standard: protein (conc : 8gm/dl)
or, Na-azide (conc : 8gm/dl)
specimen: serum
procedure: Arrange the test tube & mark then as reagent blank, standard & sample.
Pipette in cuvette sample standard reagent blank
Sample 20ml - -

Reagent 1000ml 1000ml 1000ml
Mixed & in cubate for 10 minutes at 20-25o C temperature & standard against the regent blank within 30 minutes.
Assay:

Optical pathway : 1cm
Temperature : 22-25oC
Measured against reagent blank.
Normal range: children from 3 years & adult : 6.62-8.7 gm/dl
Normal born baby : 4.6-7 gm/dl
Result: name of student : Tomal
Age : 20
serum total protion level : 11.4mg/dl
Comment: The total protein level of the subject is above normal.
Precaution:
1. Blood should be drawn with aseptic precaution.
2. Centrifugation should be completed properly.
3. 3 appropriate filter should be used.
